How does the heart use glucose and lactate at the same time? New research reveals how this system helps fuel the heart under stress.

Love this figure from our paper. Great summary here too!

Cardiac bioenergetics rewritten: MCT1 directly imports lactate into the mitochondrial inner membrane, keeping the heart alive when classical pyruvate transport fails @molecularpepsi Nice work ! 👇

🔹 MPC Bypass Uncovered: Knocking out mitochondrial pyruvate carrier (MPC) doesn't block glucose oxidation; glucose carbon enters TCA via an MCT1-dependent mitochondrial pathway.

🔹 IMM Subcellular Localization: MCT1 and LDHB reside inside the inner mitochondrial membrane/matrix, generating NADH & pyruvate locally to feed the electron transport chain directly.

🔹 Stress Vulnerability: MCT1 loss alone is compensated at baseline, but under neurohormonal stress (ATII/PE) or TAC, driving MPC suppression, MCT1 deletion drops LVEF (vs controls) & significantly increases mortality.

🔹 Translational Relevance: Human failing hearts show increased mitochondrial MCT1 localization, highlighting mitochondrial lactate import as a vital target for metabolic stress & heart failure.

Dogma-defying 😆 let’s go! I always read papers that challenged the field but never thought that I would be apart of one myself. So awesome 👏

I first presented this story at @KeystoneSymp in Breckenridge in 2022.

@ZoltArany was skeptical. But he took the idea seriously.

Over the next few years, whenever we crossed paths, he kept challenging me on it. Those conversations helped me understand just how high the burden of proof would be to convince the field. The paper is better because of them.

Eventually: “You might be right.”

Four years later, @AranyLab’s Trace Thome and Zolt have written a thoughtful commentary on our paper. Apparently we’ve advanced to “dogma-defying.”

One day I might actually be right...

These discoveries were not possible without the @RutterLab!

A tremendously collaborative effort led by co-first authors: @molecularpepsi (our amazing former postdoc), @joevisker, @jn_velascosilva, and James Carrington (our awesome grad student)!

Congrats to all authors, co-senior authors @gsducker and @StavrosDrakos, and everyone who contributed!
